He is correct that we are a circlejerk. He is also correct that we are not a "major" community.
Beyond that, there really isn't much in that statement to take seriously.
His statement "I alienated opchan by getting in an online flamewar involving one of their members" ignores that that we initially gave his comics a fairly positive reception. And then he flipped out at us posting things on the internet, that he had put on the internet; calling us hackers and demanding that we remove the images. He burned that bridge all by himself.
He points out our use of "butthurt, while himself using the words "circlejerk" and "neckbeard" to try and disparage us. That's a bit like saying "Only faggots use the word nigger
He uses sarcasm to try and deflect the bruising to his ego as seen both here "Oh boy, they must be a giant slice of possible readership" and in his comics. Sarcasm should generally be avoided in both humor and arguments, because it is typically used poorly and just makes the user sound like a grade schooler.
He claims to have decent internet popularity. If we assume that this is true, what does that even matter? Goddamn Sprite comics are popular on the internet too (yes, even after all these years). Popularity does not indicate quality, just try and suffer through and episode of Big bang theory, one of the most popular television shows.

That entire statement was basically "You are not important, it is me who is important". This is exactly the sort of thing he needs to stop doing.
But let's ignore shitty internet drama for a moment and actually glance at his comic, in a critical way.
Art: The art is shitty, but workable. This is not an insult to the artist. It just is. Two related examples that come to mind are Penny arcade and CAD, which both started out with shitty art (go and look). The guy who drew for penny arcade made a conscious effort to improve and did. Tim Buckley of CAD somehow didn't. It is my sincere hope that Mr. Grant gets better, which he should if puts any effort at all into it. As they say, draw erry day.
Writing/jokes: I didn't smile once. His joke are really lame to be brutally honest. Admittedly, it is probably much hard to impress me than it would be for most people on this board, and certainly on the internet in general. But his jokes just have a sort of "high school level" feel to them. It's the kind of stuff that I remember encountering when internet comics suddenly became a thing and every asshole with mspaint wanted to be the next big thing. This brings up another point, I've seen a decade of the worst webcomics with some that evolve to get better, and a precious few that are good right off the bat (typically because the author had considerable experience beforehand). Mr. Grant is a novice, and any critical effort should keep that in mind. Writing humor is much like art, you need to practice, but also make a deliberate effort to learn how to be better.
The biggest tip for success I could give him is to point out that comics are a visual medium. With enough artistic proficiency and subtle details, even bad jokes can be made funny, or at least interesting. The master key here is doing facial expression just the right way. Not exaggerated, but something that inherently brings out a response from an onlooker. That art by itself without any words should be funny/interesting. Comics being a visual medium also means avoid walls of text (which I fortunately have not seen him do, though sometimes he starts to get close). Comics always need a high ratio of art over words. I would also recommend another font.
All in all needs to drink some beer, say "fuck it, whatever", and forget that internet drama is even a thing (and to stop trying so hard to convince us that he doesn't care, which doesn't fool anyone but himself) because he is now the artist/writer of a webcomic. He's gonna need tougher chops, because frankly this little chan is nothing compared to the hate that will come his way from a real audience. At least he isn't using goddamn sprites.
